Can you take a photo from tumblr to walgreens/staples to have it made into a poster?
Ultimately, this question leans into the category of copyright, as I've tried to have photos turned into posters on sites like Zazzle and Redbubble, having been rejected by both for possible copyright infringement. Now, I would love to buy from the pictures, but I highly doubt that some photo someone took on an iPhone is going to be hitting the market. So, would Walgreens prevent me from printing a photo into a poster? Any help is appreciated, thanks! X

You are correct, it is a COPYRIGHT issue. Walgreens is no different than anyplace else. It is not your photo, end of discussion. No matter the source or whether it will ever "hit the market" as you say is a moot point. It is not your photo to have printed. Print sources have had to become very strict about this because they can be liable for fines of multiple thousands of dollars for unauthorized printing.
Lastly, even disregarding the copyright issue, you would be HIGHLY UNLIKELY to get a good poster size print from any photo you have taken off Tumblr. Photos on such sites are of low resolution and size. Yes, they look fine on the computer, but that is a totally different set of specs compared to what is needed for a good print, and especially a very large print such as a poster.
Why not just get yourself a good little point and shoot digital camera and learn to use it and enjoy the pleasure of taking your own photos? You then have the freedom and basic quality needed to make any prints you want.
As Steve says, it's not yours to print. That's theft.
If you want a print, contact the photographer and ask for one.
Like any other federal crime, you can certainly try -- while hoping that you are not caught. Of course, the people who run those places should have the proper training to recognize works for which you have no copyright ownership or license and they would then prefer not to get involved in being charged for a federal felony, not to mention sued when someone finds out where you got your posters made.
Of course, if you ask the copyright owners for a signed and notarized license, which you can show to a retail copier, then good for you.

You already know the answer seeing as you have tried and been refused for copyright infringement previously.
YOu need the permission of the copyright holder so ask the twitter user who originally posted the pic initially
You should not even print a copyright photo at home on your own printer, let alone try to persuade a store to break the law and make a big print.
And you might not even get a good print at 8 x 12 inches from images from Tumblr etc.
