Unless you use some third-party app, the backup of any iPhone performed with iTunes is saved on that Mac at this location:
~/Library/Application Support/MobileSync/Backup
The "~" means your home directory.
You can use one drive to backup two Macs via Time Machine. Read the long details at the article linked below.
In other words, use iTunes to backup the iPhone, and then use Time Machine to backup the two Macs.
Bear in mind that the Apple-designed processes are very easy to deal with if you must later restore the backup to a different Mac (OS X version is an important consideration). If later you have a PC (Windows), and want to get media files or documents from the backup, it can be done, but it is far from simple, because Time Machine uses a sparse bundle to isolate the files for each backup.
