If you mean restoring, then it depends on how your 3GS was unlocked: If it was unlocked using a jailbreak, the jailbreak will be lost. If it was unlocked using a proper factory unlock, the unlock will stay even after restoring via iTunes.

If it was factory unlocked like i unlocked at: http://www.iphonefort-mobile.com/ then the unlock will not be lost.
