Are you sure it's 'disk' and not iPhone is locked? Usually, when I switch off my iPhone, turn it back on but don't put in my keypass or sim-clock code and try to sync iTunes complains it needs to be unlocked first.

Your computer disk is protected in any way by any software from external writes? Also, check you anti-virus and its active protection settings - disable completely for a moment to test as it might be preventing any data from devices to be written onto your disk.

Go to the folder on your computer, find the file & make sure that the file isn't "read only", or that your'e not trying to sync it with a drive or folder that is a "read only volume."
